Pharmacology and toxicityPharmacological action
Nefopam hydrochloride is a non-narcotic central analgesic whose structure is different from other known analgesics. In vitro, nefopam hydrochloride inhibited the reuptake of catecholamine and serotonin in rat synaptosomes. In animal studies, nefopam showed analgesic properties. Nefopam hydrochloride also has antihyperalgesia effect, but its mechanism has not been fully confirmed. In clinical trials, nefopam hydrochloride has an effect on postoperative chills. Nefopam hydrochloride has no anti-inflammatory and antipyretic activity. Nefopam hydrochloride did not cause respiratory depression or reduced intestinal transport. Nefopam hydrochloride has anticholinergic activity. Nefopam hydrochloride can cause a moderate but transient increase in heart rate and blood pressure.
