pharmachologic effect
Furosemide can inhibit the reabsorption of Na and Cl by the proximal tubule, distal tubule and medullary loop of the kidney, resulting in increased excretion of water, Na and Cl and exerting a diuretic effect.
Toxicological studies
Genotoxicity:
Furosemide has no mutagenic effect on Salmonella typhimurium; However, in the presence of S9 in rat liver, high doses of furosemide can lead to suspicious positive gene mutations in mouse lymphoma cells. In in vitro assays on human cells, furosemide did not induce sister chromatid swaps, but it did induce chromosomal aberrations. In the Chinese hamster cell test, furosemide induced chromosomal damage, but the induction of sister chromatid swap was suspiciously positive. The effect of furosemide on chromosomal aberrations in mice cannot be determined. After furosemide administration to rats, their urine did not cause gene exchange in Saccharomyces cerevisiae.
